Oh my God...there's so much to break down and analyse! It starts with Thor landing on the windshield of the Milano, which is the ship of the Guardians of the Galaxy. I'm guessing that something happens and he is launched into space at the end of Thor: Ragnarok, since he is still in his gladiator outfit.
Loki is seen giving the Tesseract to someone. My guess is Thanos, since if I'm not mistaken, the Tesseract has one of the Infinity Stones. He may be a villain or anti-hero in the film.
Next, we see the hairs of Peter Parker's arm stand on end--proving that he has Spider Sense in the MCU. A few shots later, he and Tony Stark are standing alongside Mantis, Drax, and Star-Lord--and he's in the suit that he was offered at the end of Spider-Man: Homecoming. After that, the Mad Titan himself, Thanos arrives via a portal--and the battle begins.
Star-Lord is seen jumping across platforms created by Doctor Strange, and we see Spider-Man in his new suit as well. Next, we see a Wakandan army with the Winter Soldier holding a gun. They're probably heading off to fight Thanos. There is also a shot of Vision in prison (crappy rhyme intended), likely captured by Thanos since he has the Mind Stone in his forehead.
Moments later, one of the most epic shots is seen--Captain America emerging from the shadows, with a full beard. He's probably been in hiding after the events of Civil War and has reemerged to fight Thanos. I do have fears that he might die, however. Oh yeah, and the Hulkbuster suit is back, baby! We also see Black Panther in action.
After that, we see Tony with an injured Peter Parker, who says "I'm sorry, Tony." Next, we see Thor getting his head squeezed by the Mad Titan, which may hint at the God of Thunder's demise.
The trailer ends with a defeated Iron Man watching helplessly as Thanos uses the Infinity Gauntlet to unleash a hailstorm of flaming rocks upon them, and the trailer ends.
Holy sh!t that trailer was intense. I'm guessing that some big characters are going to die. The most likely ones are Vision, Iron Man, Captain America, Thor, Star-Lord, Gamora, and Nebula. Vision has the Mind Stone in his forehead, which Thanos needs for the Infinity Gauntlet. Robert Downy Jr. has hinted that he may leave the MCU, and a likely cause would be his character's death by the hands of Thanos. Cap may also die, and many have speculated on his death.Thor was screaming in pain as his head was being crushed by Thanos's grip, and Star-Lord was shown falling to the ground towards the end of the trailer. Nebula and/or Gamora may die since they are the biological daughters of the Mad Titan.
Those are my thoughts, and feel free to share yours below!

Good grief.
